AIM: This study compares the structural organization of the lung at the 20th day of embryonic development with its mature postnatal form, aiming to elucidate critical stages of pulmonary organogenesis through morphological, ultrastructural, and stereological analyses. MATRERIAL and METHOD: In this study, 10 twenty-day-old Wistar Albino rat fetuses and 10 newborn Wistar Albino rats, regardless of sex, were used. Lung tissues were dissected and examined using light and electron microscopy to evaluate developmental transitions from fetal to neonatal periods RESULTS: At the 20th day of embryonic development, the lung was found to be in the canalicular stage, characterized by immature bronchiolar structures, cuboidal epithelial lining, and a dense mesenchymal matrix. Alveolar sac formation had not yet occurred, and vascularization was incomplete. In contrast, the postnatal lung exhibited well-developed alveoli, thin septal walls, and differentiated type I and type II pneumocytes with prominent lamellar bodies, reflecting the onset of surfactant production and gas-exchange functionality. In addition to descriptive histology, stereological methods were applied to quantify developmental changes. Parameters such as total lung volume, alveolar volume fraction, septal surface area, and mean alveolar number were estimated using systematic random sampling and unbiased stereological techniques. These quantitative data provide an objective measure of the structural maturation of the respiratory parenchyma. CONCLUSION: Understanding these developmental transitions offers valuable insights into the structural and functional maturation of the lung and the origins of neonatal respiratory disorders associated with impaired alveologenesis
