Optimization of protein extraction with green chemisty approach from potato peel by the experimental designHande Konuksever1, Oğuz Akpolat2, Fatma Ayhan31Muğla University, Faculty of Science, Depatment of Chemistry, Muğla2Muğla University, Faculty of Science, Depatment of Chemistry, Physical Chemistry Department, Muğla 3Muğla University, Faculty of Science, Depatment of Chemistry, Biochemistry Department, Muğla Potato (Solanum tuberosum L.) is one of the world's most important food products, boasting high nutritional value. Therefore, potato protein holds a significant place among the proteins in food. It has been determined that potato peels are also found in significant amounts as waste around the world. In the study, the storage time, temperature, and mechanical stress were identified as important independent variable parameters for the preservation and recovery of potato protein in the extraction of potato peel protein. For this purpose, potato protein extraction was performed according to the Box-Behnken central composite design method within the specified parameter ranges (1-96 hours waiting time, 4-36°C extraction temperature, 200-600 rpm). Extract recovery yield was calculated, protein analysis was performed using the modified Lowry protein assay, and pH values were measured after extraction. The extract recovery yield, calculated based on the weights, ranged from 6.96 to 11.78 g/kg potato peel, and the percentage protein recoveries according to the modified Lowry protein analysis method ranged from 1.56 to 8.12. It was determined that the pH value decreased with extraction, and the pH values decreased with each extraction as the number of extractions increased. The results obtained from the experimental design was expressed by a mathematical responce model using a computer program. According to statistical analysis, extraction temperature is the most significant variable affecting protein extraction from potato peel. The results provide important findings within the scope of sustainable living and show that a significant waste contains compounds that can be utilized by humans.
